چکیده

We studied the effects of hyperthermia (up to 43°)on the cytolytic activity (''Cr release assay) and differentia tion of primary and secondary cytolytic T-lymphocytes (CTL) in vitro. Mixed-leukocyte cultures were established from splenic leukocytes of C57BL/6 mice and DBA/2 stimulator lymphocytes, and the CTL formed on Day 4 (primary) or on Day 21, 3 days after restimulation of the cultures with alloantigen (secondary), were assayed against P815 mastocytoma cells. Hyperthermia (43°) caused a marked decrease in cytolytic activity, the re sponse curves for primary and secondary CTL being characterized by an initial shoulder followed by an expo nential curve with an increment of dose needed to reduce activity to 37% of a starting value of 4 or 2 min, respec tively. Some of the damage was repaired when the cells were incubated at 20°or 37°for several hr (peak = 4 hr), and neither suppression of protein synthesis nor damage to membrane lipids seemed to be the causative mecha nism. Heat also affected the differentiation of precursor cells into CTL. In both short-term mixed-leukocyte cul tures stimulated with 2-mercaptoethanol and long-term mixed-leukocyte cultures stimulated with alloantigen, heating to 43°before stimulation caused a 2to 3-fold increase in subsequent CTL formation with short heating times, followed by a severe decrease with longer heating times. The results demonstrate that both primary and secondary CTL, as well as the cells Involved in their differentiation in this system, are quite sensitive to tem peratures in the range that is proposed for use in tumor therapy.

Effect of hyperthermia before Neutron irradiation on chromosomal damages of human lymphocytes

Due to importance of combination therapies in cancer treatment, this study was performed to find the effects of hyperthermia and neutron irradiation and heir combination on the human lymphocytes. Blood samples were treated separately with 41.5 °C for 30 and 60 minutes, 43 °C for 15 and 30 minutes, as well as 10 cGy neutron irradiation. They also treated with hyperthermia followed by irradiation...

Effect of LIGHT Adjuvant on Kinetics of T-Cell Responses Induced by HSV-1 DNA Immunization

Background: Studies on efficacy of various vaccines that prevent or reduce the primary and recurrent HSV-1 infection have demonstrated the importance of cellular immunity for protection against the infection. We previously used DNA vaccination to induce cellular immunity against HSV-1 infection in mice. Objective: The aim of our study was to evaluate the effect of LIGHT, a member of TNF super f...

Modification of 10 cGy neutron or gamma-rays induced chromosomal damages by hyperthermia: an in vitro study

Background: To evaluate the effects of hyperthermia (HT) on the frequency of chromosomal aberrations induced by a low dose of neutron or γ-rays in human peripheral blood lymphocytes. Materials and Methods: Blood samples were exposed to HT (41.5°C for 30 and 60min, 43°C for 15 and 30min), 10 cGy neutron or γ-rays, HT + neutron/γ, and neutron/γ + HT. After standard cell culture, harvestin...

The Influence of Perforin Expression on the Sensitivity of LAK/NK Killing Against Various Tumor Target Cells

Background: Perforin is known to be important in cytolytic activity mediated by natural killer (NK) cells.   Objective: To study the relationship between the efficiency of NK and lymphokine-activated killer (LAK) cells activity, and the expression of perforin and HLA class I molecules.   Methods: LAK cells were generated by in vitro culturing of human peripheral blood lymphocytes (PBLs) in the ...
